Monoclonal antibody (mAb) therapeutics are widely-used to treat ailments including cancer, rheumatoid arthritis, dementia and diabetes. Agarose-based chromatography resins are a vital technology for biopharmaceutical developers to obtain optimal purity in downstream processing of monoclonal antibodies and recombinant proteins.
The purification of commercially-available monoclonal antibodies (mAbs) on the market today is typically performed in three chromatography steps. Purolite offer industry-leading solutions for each step.
Protein A Affinity Chromatography
A protein A affinity resin is utilized as an initial antibody capture step. Praesto protein A resins deliver exceptionally high purity (>99%) and yield in a single, efficient step.
Cation Exchange Chromatography
A Sulphopropyl- (SP) functionalized cation exchange resin in 'bind and elute' mode is used to remove aggregates and HCP (Host Cell Proteins).
Anion Exchange Chromatography
A Quarternary Amine (Q) functionalized anion exchange resin is used in 'flow-through' mode to remove trace contaminates and ensure sufficient viral clearance.
A variety of base matrix materials can be used for mAb purification. Among these, agarose is widely considered the best material available. It is useful as it does not absorb biomolecules or their contaminants to any significant extent, has excellent flow properties, and can tolerate extreme levels of pH and ionic strength.
Agarose is a polysaccharide, generally extracted from certain types of red seaweed –Gracilaria and Gelidium. Agarose is one of the two principal components of agar and is purified by removing its other component, agaropectin. It is frequently used in molecular biology for the separation of large molecules, especially DNA, by electrophoresis. Agarose is an inherently soft material, so we use sophisticated emulsification and crosslinking technologies to increase rigidity. This ensures excellent pressure flow performance.
Agarose has a variety of characteristics that makes it ideal for antibody/protein purification, such as:
All Praesto products are manufactured with our patented Jetting technology. For faster, greener and more consistent bioprocessing.
Ecolab's goal is to secure supply, enable lot-to-lot consistency and minimize the impact of change. As such, we have made continual strategic investments to offer industry-leading security of supply and business continuity plans.
