Meeting Modern Demands of Cleaning-in-Place Protocols

As downstream purification advances, the use of enhanced cleaning-in-place (CIP) protocols has expanded to maintain product integrity and ensure high-quality biopharmaceuticals.

Purolite DurA Cycle A50 is engineered to purify monoclonal antibodies (mAbs) and associated protein constructs under variable cleaning strategies.

Compared to standard affinity resins, Purolite DurA Cycle A50 offers mild pH elution, addressing aggregation and subsequent purity challenges.

Designed to reduce susceptibility to protein A leakage, Purolite DurA Cycle A50 offers an extended resin lifetime, lowering the cost per gram.

Product Data

Dynamic Binding Capacity
Purolite DurA Cycle A50 exhibits high dynamic binding capacities for conventional monoclonal antibodies, up to 80 g/l at a 10-minute residence time. Patented Jetting technology, coupled with the latest Protein A ligand technology, allows for high capacity even at low residence times. The resin is uniquely suited to address the demands of modern bioprocessing, from high-titer cell cultures to continuous chromatography formats.
Alkaline Stability
0.1 M Sodium hydroxide (NaOH) exhibits high efficiency in removing bound proteins, nucleic acids, and lipids from bioprocess resins, alleviating the risk of fouling on heavily burdened Protein A columns. Higher concentrations of sodium hydroxide may be used between campaigns and when more substantial cleaning is required.

Purolite DurA Cycle A50 is alkaline stable for over 60 hours of exposure to 0.5 M NaOH, and over 300 hours of exposure to 0.1 M NaOH.

Milder pH Elution Window
The ligand design allows for elution at a wider range of pH when compared to conventional Protein A chromatography. Allowing for a wider operational window to address aggregation concerns and increase purity clearance within the capture step.
